The lead program, HZ01, is a RAC1 inhibitor aimed at Acute Liver Failure (ALF) and Acute-on-Chronic Liver Failure (ACLF). In preclinical studies, HZ01 reduced liver damage indicators by 40–60% across models of acute and chronic injury, suggesting potential as a pharmacological alternative to transplant.

The second candidate, HZ02, is an epigenetic reprogramming compound that promotes rapid liver regeneration. In animal models, HZ02 restored up to 67% of liver mass within 10 days following major resection and demonstrated the ability to expand healthy liver tissue while reducing tumor burden in colorectal metastasis models. Optimized formats have been developed for both oral and intravenous administration.

Spectrum’s pipeline also includes ex vivo applications, such as enlarging donor livers and recovering marginal organs, which could help expand the transplant pool.

The medical need is significant: liver diseases account for over 2 million deaths annually worldwide, with limited pharmacological solutions available. Acute and chronic liver failure, hepatocellular carcinoma, and colorectal cancer metastases to the liver remain major drivers of mortality.
