Perpetua Makwetla has a diverse work experience spanning multiple industries. Their most recent role is as a Non-executive Director at Tshwaranang Legal Advocacy Centre to End Violence Against Women, which they began in July 2021. Prior to that, they served as the Managing Director of Ketopele Human Capital Solutions starting in December 2019. Perpetua also held positions as the Head of Talent Management at JSE from September 2018 onwards, and as the Group Head of Talent and Performance Management at Motus Commercials from February to August 2018. Their previous roles include being the Country Learning and Development Manager at LafargeHolcim from August 2013 to December 2017, a Learning and Development Consultant at Trilex Group from July 2011 to July 2014, and a Learning Specialist at Edcon in 2011. Perpetua Makwetla's work experience demonstrates their expertise in talent management, learning and development, and advocacy for women's rights.

Perpetua Makwetla began their education in 2002 at the University of Johannesburg, where they pursued a Bachelor of Commerce degree in Sports Management. Perpetua completed this program in 2004. Afterwards, in 2005, they furthered their education at the same institution by completing a Bachelor of Commerce Honours degree in Sports Management. In 2009, Perpetua attended the University of Johannesburg once again, this time earning a Bachelor of Commerce Honours degree in Strategic Management. Their academic journey continued in 2018 when they enrolled at the University of the Witwatersrand to pursue a Masters in Management in Business Executive Coaching. Perpetua is currently pursuing their degree in Executive Coaching and has not yet completed their studies.
